BD FACSAria™ III Cell Sorter

BD FACSAria™ III Cell Sorter

Brief Description

The BD FACSAria™ III Cell Sorter is intended for research use and carries CE marking. It performs cell measurement, sorting, and analysis and is mounted on a dedicated mobile workstation.

Specifications

Data Management

  • Dedicated workstation
  • 262,144 analysis channels

Fluorescence Compensation

  • Unlimited inter-beam and intra-beam compensation

Sample Flow Rate

  • Analyzes up to 70,000 events/second using 8 parameters with real-time compensation of 12 spectral overlap pairs.
  • With fewer than 7 parameters and a real-time 6-color compensation matrix, sorting decision speed exceeds 100,000 events/second, with sorting performed across 260,000 analysis channels.

Cell Sorting Performance

  • Drop drive frequency: 1–100,000 Hz
  • At 70 psi and 90 kHz, with an average threshold rate of 25,000 events/second, four-way sorting achieves:
    • Purity: >98%
    • Recovery: >80%

Sorting Modes

  • Two-way sorting into:
    • 1 mL microtubes
    • 12 × 75 mm tubes
    • 15 mL tubes
  • Four-way sorting into:
    • Microtubes
    • 12 × 75 mm tubes
  • Automatic Cell Deposition Unit (ACDU) supports collection into:
    • Coverslips
    • Microscope slides
    • 6-, 24-, 48-, 96-, and 384-well plates
    • 60- and 72-well Terasaki plates

Optical Excitation

The system features a gel-coupled quartz flow cell permanently aligned with the laser optics. This design ensures optimal laser focusing on the sample stream, maximizing signal intensity and fluorescence collection. Emitted light is collected through fiber optics and directed to octagon and trigon detection assemblies, enabling simultaneous detection of fluorochromes even with identical emission wavelengths. A key innovation is the use of reflective optical filters instead of conventional transmission filters.

Light Sources / Lasers

  • Blue: 488 nm
  • Red: 633 nm
  • Violet: 405 nm
  • Yellow-Green: 561 nm

The instrument includes the necessary optical filters to detect:

  • 14 fluorochromes
  • Forward Scatter (FSC)
  • Side Scatter (SSC)

Specifically:

  • FSC is detected using a photodiode.
  • SSC is detected using a photomultiplier tube (PMT) excited by the 488 nm laser.
